Microspore Culture of Broccoli , (Brassica oleracea Var . italica)


Abstract
Culture of isolated microspores of broccoli was studied with 6 F, hybrids and 12 doubled haploids derived from anther culture of the F, variety, ¢¥Green Valiant¢¥. Three culture media reported for microspore culture in several Brassica crops and 4 microspore densities from 2.5¡¿10^4/mL to 2¡¿10^5/mL were compared. Genotypic specificity in embryo induction from microspores was very great. The F, hybrid, Green valiant, and the doubled haploid, B9, showed the highest embryogenic ability in each corresponding group. The 1/2 NLN medium was much better than the original NLN and the 1/2 NLN supplemented with BA 0.05 §·/L and NAA 0.5 §·/L. The optimum microspore density was 1¡¿10^5/mL. Since around 400 embryos were obtained from single Petri dish containing 2 mL of culture solution, the microspore culture technique was considered as a prominent method for inbred development in broccoli.
